Welcome to TomoGuide 🧊
This website serves as a resource for both beginners and experienced researchers in cryo-ET, offering an overview of the technique and data processing workflows. You’ll find practical guidance, software recommendations, and a step-by-step tutorial for preprocessing and sub-tomogram averaging (STA), along with a test dataset to practice and explore the workflow.
Start by selecting a topic from the sidebar!
You have multiple options to choose from!
- If you want to start from scratch (raw frames) using the Chlamy dataset you can start by going to Dataset and then following either one of the Scipion, RELION5, or AreTomo3 preprocessing workflows.
- If you want to start from preprocessed data (originally preprocessed in TOMOMAN) of the Chlamy dataset and learn STA using RELION5 – you can use this script to create the necessary files to start STA directly.
- If you want to use your own data, explore our Tutorial page.
We hope it will be useful!
Please keep in mind that there are dozens of cryo-ET software packages and workflows out there. What we have summarised here are common workflows we (the Cell Architecture Lab) use. Naturally, the field is constantly evolving and progressing. If you have any ideas, issues, suggestions, comments etc., please raise them directly on GitHub.
We appreciate any user feedback and are looking forward to interacting with you! Ultimately, this is all about creating a useful resource for the community. So YOU can help us make it better 😊
